Derivation of Adult Human Cortical Organotypic Slice Cultures for Coculture with Reprogrammed Neurompus slice preparation. Here we describe the derivation of human organotypic slice cultures of cortical origin, which can be kept in culture for up to 6?weeks. This method enabled us to test the system in coculture with reprogrammed neurons and show its feasibility in neuronal cell integration experiments in human-to-human grafting situation.

Direct Differentiation of Functional Neurons from Human Pluripotent Stem Cells (hPSCs),pidly derive functionally mature neurons directly from human pluripotent stem cells (hPSCs) through a brief induction of defined transcription factors. Combining Cell Fate Reprogramming and Protein Engineering to Study Transcription Factor Functions,contribution (activation or repression) of specific neuronal reprogramming factors during direct cell fate conversion. This method can be extended to every reprogramming regime to enable the functional categorization of any transcription factor.
